Neither are confirmed.

This is MadaraxSasuke talk, first part is basically explanation about Itachi. Madara talking about how Itachi was infiltrated inside clan etc. He said Konoha gave him Totsuka sword and Yata mirror.

Second part is more interesting. Madara is saying that Sasuke got unique chakra, and that he want revenge on Konoha. He's asking Sasuke to unite their chakras and have their revenge.
Truth
Madara 「Itachi took the clan upon himself, making good use of his Doutjutsus, Why you were the only one left alive? Why Itachi was the only person charged with this crime」
Sasuke: 「....」
Madara「The murder of the clan had not cooperators, not other than Itachi」....

Scene Changes

Kisame 「It looks that Sasuke won somehow... however Suigetsu, would you like to continue? it could be a little dangerous from now...」
Suigetsu 「What it is? Where is Sasuke?」
Kisame「Itachi...」
Karin 「The people of Konoha are coming to here, they might be looking for Sasuke」
Kisame 「Hurry up... I sill have work to to with Sasuke」

クーデターは九尾でやろうとしてたうちは一族はナルトが必要
The Uchiha clan, wanting to use the Kyuubi in a coup d’etat against the village, needed Naruto.

でも三代目がナルトを監視してたから（水晶で見てる）
But the Third Hokage kept an eye out on Naruto (using his crystal ball)

一族が強行手段に出るのは難儀でフガクはイタチを暗部に送ってナルトの監視役にさせようとした
The clan knew that taking Naruto by force would be difficult so Fugaku decided to send Itachi to work in the Anbu and spy on Naruto.

でも先に行動に出たのは上層部の３人でイタチに一族がクーデター起こそうとしてると話す
But the three top officials (The two people always with the Third and Danzou) got to Itachi first and told him of the clan’s plan to overthrow the Leaf.

九尾事件で子供のイタチがミコトに庇われて九尾を見てる回想九尾
Itachi has a little flashback of when he was a kid and the Kyuubi attacked (him being protected by his mother, Mikoto)

「イタチはあれを天災じゃないと聞かされて信じ込んだ最期まで信じていたようだ」「あの時天才以外に九尾を呼べるのはオレしかいなかった」「オレがイタチに嫌われている原因のひとつだった」 ."
Madara: Itachi was told that the Kyuubi attack was no natural disaster and believed it. It seems he believed it up until the very end. Back then the only person apart from a genius who could have called forth the Kyuubi was me. That’s one of the reasons Itachi hated me.

サスケ「お前とイタチはいつから繋がった」
Sasuke: So when did you and Itachi meet/join forces/etc.?

マダラ「それは後々話そう…ただ」「お前がイタチを兄と慕っていた時間より長いとだけ言っておく」
Madara: That conversation can wait till later…However, I will say now that we knew each other longer than you thought of him as your beloved older brother.

サスケ「…」
Sasuke: …

イタチには考える時間が与えられた
Itachi was given some time to think.

イタチは悩んだ…一族を皆殺しにするという事はサスケ、もちろんお前もその対象に入っていた
Itachi was troubled…killing his entire clan would mean that Sasuke…of course you (Sasuke) would have been included.

だが板挟みになって身動きが取れなくなっていたイタチを決意させる決定的な出来事が二つ起こる
But for Itachi, whose dilemma had left him unable to act at all, two things occurred which would help him make his decision.

ひとつはシスイ殺しの疑いをかけられた事だ
The first was the suspicion put on him that he had murdered Shisui.

強い憎しみからイタチは万華鏡写輪眼を開眼した
Itachi awoke the Mangekyou Sharingan from an intense hatred.

サスケ回想「あの時…！」
Sasuke flashback: That time…!

だがそれはフガクが仕向けた事だった. 九尾を操るには万華鏡写輪眼が必要だ
But that was what Fugaku had manipulated him into. The Mangekyou Sharingan was necessary in order to control the Kyuubi.

イタチの様子がおかしいと感付いていたフガクは可能性があったイタチに万華鏡を開眼させることでイタチを一族に縛りつけようとした
Fugaku, sensing that Itachi was acting strangely, decided to awaken the Mangekyou Sharingan in Itachi, as he had the possibility of attaining it, and using this to tie Itachi to the clan.

サスケ「父さんが…」
Sasuke: Father…

そしてもうひとつは里の上層部からお前の命を引き合いに出された事だ
The other thing that happened was that three higher ups threw your (Sasuke’s) life into the mix.

じいさんばあさんがクーデターが起きたら関係ない人が死ぬ恐れもあるし
The old man and lady thought that were the coup d’etat to happen that many people unrelated to the happenings might die.

どっちみち一族も殺すことになるから
If either way the clan is going to be killed

サスケだけでも助けたくないかと言われてうなずくイタチで終
Then wouldn’t Itachi want to at least save Sasuke? The chapter ends with Itachi nodding yes.

So he had no choice but obliterating the clan

The Third tried to compromise, but there was no time before that night

抜け忍になるのも指令の一つだった
Becoming a missing-nin was also his instructions
